Is it possible to take a sample of water into a jar and put it into a closet (or somewhere dark) for 8 hours to simulate an OCLT. Would the results tell you anything?
 
Only if you also scoop up some algae. Its unlikely that the algae will be floating around for you to scoop, and if it is you probably don't need a test.
The test is meant to help you determine if there is algae somewhere you cant see it, such as on ladders or behind light fixtures, etc.

No. You need to have the pool water be in the pool environment to get the impact of everything that is in the pool consuming FC.
