Always run the pump for at least an hour before you test and an hour after you add any chemicals.

I just spent a week clearing up my pool and want to make sure that I test it properly to make sure I don't ever have to wake up to a green swamp again.

Does it matter whether I run the pump at night? I currently run it for 5 hours between 11 p.m. and 4 a.m.

Does not really make a lot of difference when you run the pump. There are slight advantages running in the day, but not worth the extra cost if electricity is cheapest at night.
