It's not so much the amount of sand but the pressure you want to focus on. See the link below.
HINT: If you are just starting a SLAM Process, don't bother with the DE yet. Save that for the very last if you have residual water clarity issues.

Add DE to a Sand Filter
A clean pool filter will increase the efficiency of your pool! ... your filter are to backwash it, clean the manifolds and grids, and add new DE.
