Water sample depth

Is there really a significant difference in test readings from different water depths? If so, which variables are most affected and by how much? For instance, a test sample taken from an 8 inch depth compared to an 18 inch depth? This assumes the pump is running, I'd never take a sample without the pump running.
 
Pump running, all the chemistry should be well mixed and you, in theory, can take the sample at any depth.

Just to reduce any surface contamination, TFP suggest sampling from a foot below the surface.

The important take away is, just like you already do, make sure your pump circulates the water for maybe 30 minutes before sampling.
