Too soon to test CYA?

I used PoolMath to calculate how much CYA was needed to raise it from 30 to 50 and it called for 28oz of stablizer by volume. I added 3 cups of grandular stabilizer in a sock in the skimmer on Friday and last night the sock was empty so I removed it. Tested the CYA this morning and it was only 40. Is it likely to test higher if I retest this Friday even though there is no more in the skimmer to dissolve?
