Just added 1080 gallons of fill. Should I wait a few hours to retest considering or just test now?

I don't necessarily have an autofill. I have a line running to the pool, but I have to manually turn it on/off.

Here are my FILL Water readings CL: < .5 pH: 7.0 - 7.1 TA: 300 CH: 290
