Just tested CYA in pool to see what my target FC level should be. My test came back at CYA=80. Now here is my question pool is only in its second season ( filled last year around mid June ) and used city water to fill, so could have using pucks for just over a year bring my CYA level up so high?

Easily, especially if you do not backwash out much water or lower it to close in the winter or get a lot of rain.
How many pucks have you used? poolcalculator.com in the Effect of Adding chemicals section at the bottom will tell you how much CYA was added.

Yes they will. One 7 oz puck will raise CYA by 6 ppm in 5000 gal.
