Last year my pump seized over the winter, I managed to free it by turning the spindle by hand and it has been fine all summer. To prevent the same thing from happening, should I fill in the pump with antifreeze solution?

No. The area that would seize shouldn't be in contact with the water. Just drain the pump completely and leave the plugs out.
That won't help. Seizing is almost always related to the bearings, not where the anti-freeze could go.
That's what I do normally and previous years it's been ok until last winter when the pump seized. Should I cover the pump with some kind of tarp to keep the snow and ice away?
I don't recommend a plastic bag, but use something that will breath. You don't want to trap the moisture in the bearings.
